It wouldn't take much. Native Americans have a genetic intolerance to alcohol.
They didn't have to respond to unclean water the way Europeans did; by drinking fermented drinks instead. Most of the population in Europe developed their alcohol tolerance this way, and the ones that didn't died of disease. The ones with a low tolerance presumably died of alcohol related diseases too. You're then left with a gene pool of alcohol tolerant people.
